IncuCyte™ Plus

IncuCyte Plus is the original phase-contrast imaging system. It is configured to fit “inside” a CO2 incubator and provides only phase-contrast imaging, either via standard phase-contrast optics or via the proprietary HD Optics. This platform is useful for non-invasively assessing live cell cultures and incorporates cell confluence as its primary quantitative metric.

IncuCyte Plus is used to collect phase contrast images over time. Kinetic plots of confluence are then used to objectively and non-invasively quantify cell growth. (Click image to enlarge)

Applications

  • Cell culture quality control
  • Monitoring cell growth rates
  • Documenting cell morphology in time
  • Cell-based assay optimization

IncuCyte™ Plus provides a non-invasive and labor-free method to document your cell culture. Shown are a transfected CHO cell line at passage 3 and passage 18. Long term changes in morphology are easy to track with IncuCyte.

Example Assay: Automated Scratch Wound

One of the first applications developed on the IncuCyte™ platform was a kinetic, non-labeled, automated version of the scratch wound assay. Scratch wound assays are commonly used to assess the effects of drugs and drug candidates on the cellular proliferation and/or migration associated with wound closing.

In order to run this assay on IncuCyte™, we currently sell a 24-well plate-based scratch wound kit which includes: a.) a wound-making tool and b.) 24-well Essen Imagelock™ plates. These tools in combination insure that one achieves consistent wounds of the cell monolayer, and that the IncuCyte™ software can find and automatically quantify wound closure. A 96-well version of this assay is in development.

IncuCyte software computes wound metrics directly from HD-phase imagery. Shown are the initial wound mask (black) and the effective wound area (grey) for each respective time point. (Click to Enlarge)
