Seahorse XF Palmitate-BSA FAO Substrate

The Seahorse XF Palmitate-BSA FAO Substrate is a preconjugated, quality-controlled, fatty acid substrate for in vitro studies of long chain fatty acid oxidation by intact cells. This bioavailable, conjugated fatty acid substrate frees you from the tedious, time-consuming, and variable conjugation process. Best used for assays in which the cells are partially or completely dependent on exogenous LCFAs for maximal respiration and responses to stress.

Features

  • Ready-to-use substrate for consistent results in fatty acid oxidation (FAO) assays.
  • Free fatty acid concentration provided with each lot.
  • Reliable, functional information without radioactivity.
  • FAO assay workflow integration with the Seahorse XF Cell Mito Stress Test.

How it works

Investigate how cells oxidize LCFAs (Long Chain Fatty Acids)

Fatty acid is a major fuel that can provide up to 80% of the ATP required in some tissues (e.g., liver, heart).  Therefore, examination of fatty acid oxidation (FAO) is often a part of the general characterization of mitochondrial function.
This bio-available substrate is intended for in vitro studies of FAO by intact cells. It is commonly used with Etomoxir and XF Cell Mito Stress Test kit, and enables to investigate how LCFA oxidation responds to drug exposure and a genetic manipulation.

Examine fatty acid impact on fundamental respiration parameters

In assays with Palmitate-BSA, cells are first conditioned in nutrient-limited media, then treated with or without Etomoxir, followed by sequential injection of respiration modulators in the presence or absence of Palmitate-BSA. As a result, 4 OCR profiles are obtained and by comparing these profiles, the contribution of endogenous and exogenous fatty acid to overall mitochondrial respiration can be determined.
Etomoxir can be replaced by any compounds of interest to assess the compound effect.
